Harzianopyridone is a fungal pyridone natural product that inhibits mitochondrial complex II and Zika virus replication.
Details
Harzianopyridone is a pentasubstituted pyridone natural product produced by Trichoderma harzianum and related fungi. Biosynthetic studies revealed an iterative pathway involving methyltransferase and flavin-dependent monooxygenase enzymes closely related to the atpenin biosynthetic system. Harzianopyridone functions as a potent inhibitor of mitochondrial complex II (succinate dehydrogenase), thereby interfering with the respiratory chain. In addition, it has been reported to inhibit Zika virus replication by directly binding to and suppressing RNA-dependent RNA polymerase (RdRp) activity. These findings indicate that harzianopyridone possesses dual bioactivity targeting both mitochondrial respiration and viral RNA synthesis.
